Measurement of anti‑phospholipase A2 receptor antibodies based on surface‑enhanced Raman spectroscopy

摘要: 利用自行研制的表面增强拉曼光谱 (SERS) 测试平台, 以自制的纳米银胶体溶液为增强基底, 对不同滴度下 的抗磷脂酶A2受体 (anti-phospholipase A2 receptor, anti-PLA2R) 抗体水溶液进行检测, 发现在纳米银胶体溶液增强 作用下, 不同滴度的抗 PLA2R 抗体水溶液均能够检测出拉曼频移约为 1521 cm−1的特征峰, 并且检测下限可至 2 RU/mL。在此基础上, 进一步对健康人尿液样本和混合抗PLA2R抗体水溶液的尿液样本进行检测分析, 发现只有 在混合抗 PLA2R 抗体水溶液的样本中才能检测到 1521 cm−1 的拉曼特征峰。本研究表明, SERS 技术可以检测到 PLA2R的拉曼特征峰, 基于尿液的SERS检测有望发展成为临床无创诊断特发性膜性肾病的新方法, 对特发性膜性 肾病疑似患者是否需要进行详细的肾组织活检提供了重要的指导依据。

Abstract: The self-developed Raman spectrum test platform was utilized to detect anti-phospholipase A2 receptor (PLA2R) antibody aqueous solutions with varying titers, employing a self-made nano silver colloidal solution as the enhancement medium. It was observed that, under the enhanced effect of the nano silver colloidal solution, PLA2R antibody aqueous solutions with different titers exhibited a characteristic peak at a Raman frequency shift of approximately 1521 cm-1, and the detection limit could reach 2 RU/mL. Based on this finding, further analysis and detection were conducted on urine samples from healthy individuals and mixed PLA2R antibody aqueous solutions. Notably, only in the samples containing mixed PLA2R antibody aqueous solutions could the Raman characteristic peak at 1521 cm-1 be detected. This experiment demonstrated that surface-enhanced Raman spectroscopy (SERS) can effectively detect very low titer PLA2R antibody aqueous solutions. Moreover, SERS-based urine detection holds promising potential as a novel method for clinically non-invasive diagnosis of idiopathic membranous nephropathy and serves as an important guiding principle for determining whether suspected patients require detailed kidney tissue biopsy.
